Jerktonium only appears in It's a SpongeBob Christmas!. It is a dangerous chemical element on the periodic table discovered by Sheldon J. Plankton (no other element has been on the periodic table before). It also serves as a trap for the residents in Bikini Bottom (except for SpongeBob, of course).

Plankton uses Jerktonium in the fruitcake that is given out to everybody in Bikini Bottom. The Jerktonium causes all Bikini Bottomites to become grumpy and act like jerks in order to make Plankton appear to be the nicest of them all. Everyone who tries the fruitcake gains a mass of black stubble (a "five o' clock shadow") on their mouths and acts like a jerk to everyone else. Plankton's first victim is SpongeBob, but SpongeBob is immune to Jerktonium because of his pure heart and tiny brain, causing Plankton's plan to prove insufficient yet again. Soon Plankton gets tired and allows SpongeBob to deliver the fruitcake with his latest invention, the JerkMaker 9000.

SpongeBob goes downtown and begins passing out fruitcake while singing, causing everybody to get infected with Jerktonium and become jerks, though he is oblivious throughout and just keeps singing. At Sandy's Treedome, Sandy scans SpongeBob, who had just said that he must be the biggest jerk in town because he ate a lot of fruitcake, and tells him that everyone knows he is immune to Jerktonium. She uses the analyzer to reveal a formula for an antidote; the antidote is a song which SpongeBob hums, and it cures Sandy.

Returning downtown, SpongeBob begins singing to rid everyone of the Jerktonium, curing them. This ruins Plankton's already-insufficient plan, but Santa reveals that everyone besides Plankton is on the naughty list, calling SpongeBob the worst of all; Plankton had, in fact, made ToyBob, a robotic SpongeBob created to ruin SpongeBob's reputation. ToyBob tries to destroy Santa, but in the end it itself is destroyed by SpongeBob.
